چکیده

A pyridine nucleotide has been synthesized from cr-nicotinamide adenine dinucleotide and adenosine triphosphate with the use of a crude enzyme preparation from Azotobacfer uinelandii strain 0. Evidence is presented that this nucleotide is the (Y isomer of NADP, ar-NADP, which differs from &NADP in the configuration of the nicotinamide-ribosidic linkage. This evidence rests on a study of (a) spectral properties, (b) degradation by snake venom phosphodiesterase followed by comparison with authentic samples, (c) stability of the nicotinamide-ribonucleosidic linkage in acid, and (d) optical rotatory dispersion. In contrast to P-NADP, ar-NADP is not active with yeast glucose 6-phosphate dehydrogenase, nor is it cleaved by Neurospora crassa NAD glycohydrolase. o-NADP has been shown to be present in the cells of A. vinelandii; about 1: 10 has been found for a normal equilibrium molar ratio of a-NADP to /3-NADP.

Verifying of Participation of Nitric Oxide in Morphine Place Conditioning in the Rat Medial Septum Using Nicotinamide Adenine Dinucleotide Phosphate-Diaphorase (NADPH-d)

Background: Role of nitric oxide (NO) in morphine-induced conditioned place preference (CPP) has already been proposed in the rat medial septum (MS), but no molecular evidence has been provided to clear this fact. Methods: Effects of intraseptal injections of L-arginine and/or NG-nitro-L-arginine methyl ester (L-NAME) on morphine place conditioning in Wistar rats were examined. Morphine (2.5-7....

Biochemical and quantitative histochemical study of reduced pyridine nucleotide dehydrogenation by human colonic carcinomas.

This study shows a marked increase in the activity of the soluble enzyme DT-diaphorase and of the histochemical activity of the reduced nicotinamide adenine dinucleotide and reduced nicotinamide adenine dinucleotide phosphate nitroblue tetrazolium menadione-mediated reductases in human colonic carcinomas when compared with the enzymatic activities of portions of the colon uninvolved by the carc...

REASSOCIATION AND REACTIVATION OF GLUCOSE 6-PHOSPHATE DEHYDROGENASE FROM STREPTOMYCES AUREOFACIENS AFTER DENATURATION BY 6 M UREA

Glucose 6-phosphate dehydrogenase (G6PD) from Streptomyces aureofaciens was purified and denatured in 6 M urea. Denaturation led to complete dissociation of the enzyme into its inactive monomers, 98% loss of the enzyme activity, about 30% decrease in the protein fluorescence and a 10 nm red shift in the emission maximum. Dilution of urea-denatured enzyme resulted in regaining of the enzyme acti...

Reversible enzymatic synthesis of flavin-adenine dinucleotide.

An enzyme has been partially purified from brewer's yeast which catalyzes the reversible reaction between riboflavin phosphate and adenosine triphosphate (ATP) on the one hand and flavin-adenine dinucleotide (FAD) and inorganic pyrophosphate on the other in the presence of magnesium ions. ATP could not be replaced by adenosine diphosphate or adenosine-5-phosphate, and in the reverse reaction, i...
